A few cents off is not always audible on one string, but it compounds fast.

A slightly flat or sharp violin is easy to miss alone but stands out immediately next to a piano, another string player, or a backing track tuned to concert pitch.
Two strings sounded together make tuning errors obvious as beating or a harsh, unclear sound. Well-tuned strings ring clean and let the instrument's natural resonance come through.

Two words, two directions on the peg.
A sharp reading means your string is vibrating faster than the target frequency, so the pitch sounds higher than it should. The cents number is positive, and the needle sits to the right of center. Loosen the peg or fine tuner slightly to bring it down.
A flat reading means your string is vibrating slower than the target frequency, so the pitch sounds lower than it should. The cents number is negative, and the needle sits to the left of center. Tighten the peg or fine tuner slightly to bring it up.
A standard violin is tuned in fifths to G3 (196.00 Hz), D4 (293.66 Hz), A4 (440.00 Hz) and E5 (659.25 Hz), from the lowest and thickest string to the highest and thinnest. These values assume the standard concert pitch of A4 = 440 Hz.

A cent is one hundredth of a semitone. The readout shows how many cents above or below the target string frequency your note currently sits. A reading within about 5 cents is generally considered in tune to the ear.
Large adjustments are easier with the pegs; fine tuners on the tailpiece are better for small corrections once you are close. If the reading shows Sharp, loosen slightly; if it shows Flat, tighten slightly, then re-check because strings settle after each turn.
Yes. Auto Detect mode compares whatever pitch it hears against all four open strings and automatically targets the closest one, so you can move from G to D to A to E without changing any setting.
Manual selection is useful when a string is badly out of tune, because a very flat or sharp note can sometimes sit closer to a neighboring string than to its own target. Locking the string you intend to tune removes that ambiguity.
This can happen in Auto Detect mode if a string is tuned far enough off pitch that it lands closer to a different open string, or if a sympathetic overtone from another string is being picked up. Switch to Manual mode and select A directly to avoid this.

440 Hz is the standard concert pitch used by most schools, method books and recordings, and is the default here. Some orchestras tune slightly higher, around 442 Hz. Use the reference pitch slider to match whoever you are playing with.
New strings stretch and settle over the first days or weeks of play. Temperature and humidity changes also shrink or expand the wood and strings, and pegs can slip if they are not seated well. Frequent small re-tunings are normal, especially for newer strings.
